We%26#39;re traveling to Austria next summer. When we leave Salzburg, we%26#39;d like to drive along the Grossglockner strasse. Can we drive from Salzburg to Heiligenblut, then back to Zell am See for overnight?

You probably can do it, but by the time you lie down in your bed, your head may still be taking hairpin curves.
It would be better to spend the nioght in Zell am See and then do the trip on the next day, early in the morning and when you are fresh. You really never know how long the trip will last, because you might be caught for quite a stretch behind some slow moving bus (of which there are many!). That%26#39;s why early in the morning is the best time to start.
